Cardiotoxic effects of angiogenesis inhibitors

چکیده

Abstract The development of new therapies for cancer has led to dramatic improvements in survivorship. Angiogenesis inhibitors represent one such advancement, revolutionising treatment a wide range malignancies. However, these drugs are associated with cardiovascular toxicities which can impact optimal the short-term and may lead increased morbidity mortality longer term. Vascular endothelial growth factor (VEGFIs) hypertension, left ventricular systolic dysfunction (LVSD) heart failure as well arterial venous thromboembolism, QTc interval prolongation arrhythmia. mechanisms behind VEGFI-associated LVSD likely involve combination number myocardial insults. These include direct effects, secondary toxicity via coronary or peripheral vascular damage. Cardiac result from ‘on-target’ effects VEGF inhibition ‘off-target’ resulting other tyrosine kinases. Similar be involved right (RV) dysfunction. Some VEGFIs an risk atrial Further pre-clinical clinical studies trials needed better understand VEGFI on system. Once elucidated, investigated surveillance strategies identifying complications developed.

Cardiotoxic Effects

Renal insufficiency is regularly accompanied by an increase of the total catecholamines of the blood. Adrenolytic drugs, which inactivate circulating catecholamines, temporarily improve the abnormal S-T segments and T waves in patients with renal insufficiency but not in hypertensive patients without renal complications. It is suggested that in renal insufficiency, the retained circulating cate...

Cardiotoxic effects of trastuzumab.

BACKGROUND Breast cancer with overexpression of the HER2 receptor is an aggressive type of breast cancer with poor prognosis. Trastuzumab (Herceptin) is a monoclonal antibody that binds to the HER2 receptor on the cell surface blocking the signals that promote cell-growth proliferation. Trastuzumab treatment has almost halved the risk of relapse, when given as an adjuvant, and has improved the ...

Endogenous inhibitors of angiogenesis.

Angiogenesis, the formation of new blood vessels, is required for many pathologic processes, including invasive tumor growth as well as physiologic organ/tissue maintenance. Angiogenesis during development and adulthood is likely regulated by a balance between endogenous proangiogenic and antiangiogenic factors. It is speculated that tumor growth requires disruption of such balance; thus, the a...
